چکیده
Thank you for submitting your manuscript for consideration by the EMBO Journal. First of all, please let me apologise for the length of time taken to get back to you with a decision this was due firstly to some difficulty in finding three appropriate referees, and secondly to a delay in receiving the final report. However, your manuscript has now been seen by three referees whose comments are enclosed. As you will see, referees 1 and 3 express interest in your study and are broadly positive, although do highlight a number of concerns that would need to be addressed before we could consider publication. On the other hand, referee 2 (whose opinion is stated more explicitly in his/her confidential comments to the editor) finds that it does not make a significant conceptual advance beyond previous work (from your lab and others) that already suggested that EV1 binds to the inactive conformation of the integrin receptor.

Calpains promote α2β1 integrin turnover in nonrecycling integrin pathway
Collagen receptor integrins recycle between the plasma membrane and endosomes and facilitate formation and turnover of focal adhesions. In contrast, clustering of α2β1 integrin with antibodies or the human pathogen echovirus 1 (EV1) causes redistribution of α2 integrin to perinuclear multivesicular bodies, α2-MVBs. We show here that the internalized clustered α2 integrin remains in α2-MVBs and ...
متن کاملCholesterol Dependence of Collagen and Echovirus 1 Trafficking along the Novel α2β1 Integrin Internalization Pathway
We have previously shown that soluble collagen and a human pathogen, echovirus 1 (EV1) cluster α2β1 integrin on the plasma membrane and cause their internalization into cytoplasmic endosomes. Here we show that cholesterol plays a major role not only in the uptake of α2β1 integrin and its ligands but also in the formation of α2 integrin-specific multivesicular bodies (α2-MVBs) and virus infectio...
متن کاملClustering induces a lateral redistribution of α2β1 integrin from membrane rafts to caveolae and subsequent PKC-dependent internalization
Integrin α2β1 mediates the binding of several epithelial and mesenchymal cell types to collagen. The composition of the surrounding plasma membrane, especially caveolin-1 and cholesterol containing membrane structures called caveolae, may be important to integrin signaling. On cell surface α2β1 integrin was located in the raft like membrane domain, rich in GPI-anchored proteins, rather than in ...

متن کاملUp-Regulation of Integrinsn α2β1 and α3β1 Expression in Human Foreskin Fibroblast Cells after In-Vitro Infection with Herpes Simplex Virus Type 1
The interaction of Herpes Simplex Virus type 1 (HSV-1) with human fetal foreskin fibroblast (HFFF) cell was studied using a recent isolate of HSV-1 which was propagated in Hep-2 cells. HFFF cells were challenged with HSV-1 with a multiplicity of infection (MOI) of 1 virus/cell for 24 hours. Flow cytometric analysis demonstrated that HSV-1 challenged HFFF cells expressed increased levels of α2β1...
